胆汁酸的遗传毒性和对细胞间隙连接通讯( GJIC) 抑制作用的研究

摘要: 本文应用Ames 试验,CHL 细胞体外染色体畸变试验检测了脱氧胆酸和石胆酸的遗传毒性;还观察了脱氧胆酸和石胆酸对NIH/ 3T3 细胞的恶性转化作用。并应用划痕标记染料示踪技术(SLDT) 观察了胆汁酸对细胞间隙连接通讯( GJ IC) 的抑制作用。结果表明,脱氧胆酸和石胆酸无诱发基因突变和染色体畸变作用,也无诱发培养细胞的恶性转化作用。因此本实验未证明脱氧胆酸和石胆酸是遗传毒性致癌物。然而,脱氧胆酸和石胆酸对NIH/ 3T3 细胞GJ IC 均有明显抑制作用,说明胆汁酸在大肠癌的发生中是一种促癌剂。

Abstract: The genotoxicities of Deoxycholic acid and Lithocholtic acid were tested by Ames test and chromosome aberration test in vit ro. The malignent transformation effect of Deoxycholic acid and Lithocholic acid on NIH/ 3T3 cells was observed , The inhibitory effect of Deoxycholic acid and Lithocholic acid on gap junction intercellular communication ( GJ IC) in NIH/ 3T3 cells was investigated with Scrap Loading/ Dye Transfer test (SLDT) . The result s indicated that neither Deoxycholic acid nor Lithocholic acid has gene mutation , chromosome aberration and cell malignant t ransformation in vit ro , Therefore , it did not demonst rate that bile acid is a genotoxic carcinogen. But result showed that both Deoxycholic acid and Lithocholic acid could inhibit NIH/ 3T3 cells GJ IC obviously. It means that bile acid may play a role as promoter in carcinogenesis.
